As for content, more villains should be included in the playable roster and no more characters that appear (too) occasionally in Mario games. Or if they must bring some new characters from the franchise, they should replace (Luigi particularly). There's already too many of them, eight as of now.

Speaking of replacing, semi-clones NEED to go. Practically every semi-clone are better than their originals, it's like Street Fighter's Shoto-syndrome. We don't need two fighters *series* with near-identical movesets. Wolf, you have a chance to set things right. As well as any other semi-clone for that matter.
They could just make the clones even more different so they're no so clone-y. If you remove the clone-y people though you're losing maybe a quarter the crowd... and I'd say that the special villians should be made into bosses if they come up with a new adventure mode. many that come straight to mind would be a Kirby vs Marx section, a Metaknight vs Galacta knight section, a fire emblem cast vs Black knight section and Samus vs Dark samus section. hmmm, that makes me think... Perhaps the adventure mode could be something which is "Closer to home" which ends with everyone all together or something...
I'd like for Final Smashes to return, but I'd rather have more single-use blasts instead of ever-lasting overpowered forms. Using forms takes zero effort to get any decent results. Try kill with a Mario Finale, then do the same with a Landmaster and you'll see what I mean.
Mah boi, you're correct. They all need to be made into "moves" or harmable transformations...Harmable I say because although bowser gets giga and super armor you can still hurt him somewhat. cause when I vs someone going giga I actually take it as a chance to deal damage to him cause he takes that boss flinchy hitstun type stuff which makes him pause all the time. Transformations like Warioman... no. Broken, same with landmasters. Some of them seem legit but when I say some, I mean some... one which primarily comes to mind which is broken beyond heck is "NOW I'LL SHOW YOU! HAAAAAAAAA!" *everyone on screen dies cause it's a level 9 computer using it. T.T*. They should maybe make it like an astral finish or distortion drive thing. one use per round, but a single hit rapes the shit out of whoever it hits. Marths final smash would be perfect for an example of this kinda thing. -_-'
Third party characters. I won't mind them, as long as they are reasonable picks, like Megaman. (DO WANT CRASH THOUGH.)

But until a time for a next Smash Bros. -game comes (shall it ever be made), we could always feast on this.
